Product Description
Angiotensin II (Ang II) is the most potent pressure agent known to date and is the biologically active peptide of the renin-angiotensin system (RAS). The proteolytic enzyme renin, synthesized, stored and secreted primarily by the juxtaglomerular cells of the kidneys, cleaves angiotensinogen to produce the decapeptide angiotensin I (Ang I) which is physiologically inactive. Ang I is rapidly cleaved by the angiotensin converting enzyme (ACE) in plasma, vascular endothelial cells, kidney, lung and other tissues, into Ang II, the major active component of the renin-angiotensin cascade10, 11 (see fig.1). This octapeptide is then rapidly degraded by various angiotensinases (aminopeptidases) found in plasma and tissues to the heptapeptide Ang III and to its subsequent degradation products, the hexa-, penta- and tetrapeptides, 5,6,12,13 This assay measures Ang II by a double-antibody radioimmunoassay using a modification of the method by Emanuel et al. 1. Standards, controls and reversed phase-extracted plasma samples are incubated for 16 hours with anti-angiotensin II antibody; 125I-angiotensin II then competes with angiotensin II present in samples and standards for the same antibody binding sites. After a second 6-hour incubation, the solid-phase second antibody is added to the mixture, antibody-bound fraction is precipitated and counted.
