| Acid-labile subunit (ALS) is a GH dependent protein which is produced in the liver as a 578 amino acid peptide with a predicted molecular weight of 63.3 kDa. IGFs in human serum are mostly confined to a ~150 KDa GH-dependent ternary protein complex consisting of IGFBP-3, a molecule of IGF-I or IGF-II, and this unique acid-labile subunit (ALS) [1-6]. The first description of ALS was based on the observation that acidification would result in dissociation of the 150 KDa complex into acid-stable IGF (~7.5 KDa) and IGFBP-3 (~50 KDa) components while neutralization of the acidified sample would no longer support formation of the 150 KDa complex [7,8]. The existence of ALS was later confirmed by the finding that serum-derived ALS could readily convert binary IGF/IGFBP-3 complexes to the 150 KDa form and that IGFBP-3 occupancy by IGF-I or IGF-II was a prerequisite for the complex assembly [9,10] in vivo. However, ALS may also be capable of IGF-independent interaction with IGFBP-3 and as such modulate the binding capacity of IGFBP-3 for the IGF peptides [11].
